Purification of Glycosaminoglycan Extracted from Octopus dollfusi and Effect on Proliferation of Mouse Splenocytes

Abstract:

In this paper , the crude glycosaminoglycan extracted from Oc.ωp旧dollfusi (COG) was refined to obtain POG, and was then purified by means of Anion-Exchange and Gel Filtration Chromatography. Mterwards , chemistrγassay and IR spectrography were adopted to determine the main compositions and structures of the products , and MIT assay was used to measure the splenocyte proliferation. The results show that the total sugar contents of POGⅠ, POG Ⅱ and POGⅠ -1 purified from the chromatography are all in the range of 26.3% -42.2% , and that there is more than 30% of total glycosaminoglycan and approximately 20% of glucuronic acid. Moreover , the IR spectrum reveals that there are typical glycosaminoglycan structure characteristics , and that COG, POG, POGⅠ, POGⅡ, POG Ⅰ -1 all significantly promote the proliferation of mouse splenocytes, especial1y POG Ⅰ -1 that has the greatest effect. It thus concluded that the glycosaminoglycans from Octopus dollfusi consisting of hexosamine and glucuronic acid have the function of immunity enhancement.
