Cellular Immunity Induced by gD2 DNA Vaccine

Abstract:  pcDNA3-gD‚the constructed Genital Herpes gD DNA vaccine (gD2DNA vaccine)‚was transfected into mammalian cell COS-7‚and the expression of gD gene was investigated.BALB/c mice were then immunized by pcDNA3-gD.The proliferation of spleen cell‚the activity of cytotoxic T cell (CTL) and CD4+ and CD8 +populations were respectively analyzed.The results show that pcDNA3-gD plasmid can express the gene of gD in cell COS-7‚that the spleen cell of mice immunized by pcDNA3-gD plasmid is of an increased proliferation activity‚that the activity of CTL in the experiment group (immunized by pcDNA3-gD plasmid) is37.69%which is obviously higher than that of the control group (immunized by pcDNA3plasmid) and the naive group (immunized by normal saline)‚and that the percentage of CD4+cells in the experiment group increases to33.38% which is greater than that of the control group and the naive group.
