何首乌中Ⅲ型PKS基因的克隆和表达

Cloning and Expression of Type Ⅲ Polyketide Synthase Gene in Fallopia multiflora

摘要: 为利用基因工程技术调控何首乌的次生代谢途径,采用互补DNA末端快速扩增技术以及生物信息学方法,从传统中药材何首乌中克隆到一种Ⅲ型PKS基因FmPKS2(GenBank登录号:GQ984139),其互补DNA全长1498bp,编码392个氨基酸.生物信息学预测其蛋白相对分子质量为43160,等电点为5.85,亲水系数为-0.154,含有几个较强的疏水区域.系统进化树显示,FmPKS2与蓼科植物的查尔酮聚合酶聚类关系最近.该基因在何首乌不同组织中均有表达,其中在块根中表达量最高.

Abstract:

In order to regulate the secondary metabolic pathway of a Chinese traditional medicinal material Fallopia multiflora by genetic engineering technology,a gene named FmPKS2(GenBank accession number: GQ984139) was cloned via the rapid amplification of complementary DNA end and bioinformatics analysis.The gene consists of 1498bp nucleotides for coding sequence,encodes 392 amino acids with a predicted relative molecular mass of 43160 and an isoelectric point of 5.85,and contains several highly hydrophobic regions with a hydrophobic coefficient of-0.154.Phylogenetic analysis reveals that FmPKS2 shares the closed relationship with chalcone synthase of polygonaceae family.The transcription of FmPKS2 can be found in all different tissues of Fallopia multiflora,with the highest quantity in rhizomes.
