Preparation and Fusion of the Strain Protoplast for the Degradation of Polyvinyl Alcohol

摘要: 从自然界中分离得到4株能降解聚乙烯醇(PVA)的细菌‚经紫外线诱变‚得到两株具有单重抗药性的突变菌株S7(Str r ‚Kan s )和 K15(Str s ‚Kan r ).系统分析了各种因素对原生质体制备率以及再生率的影响;将S7和 K15作为亲本菌株进行原生质体融合‚并通过正交试验‚对原生质体融合的条件进行优化‚使融合率达到4.603×10-5 .融合子F4菌株培养成活性污泥后‚PVA 降解率可达87%‚是普通活性污泥的3~4倍.

Abstract: Four strains capable of degrading polyvinyl alcohol (PVA) were separated from natural specimens‚and two mutants‚S7(Str r ‚Kan s ) and K15(Str s ‚Kan r )‚which are of single drug resistance‚were obtained via UV-irradia-tion.The effects of various factors on the preparation and the regeneration rates of the protoplast were systematically ana-lyzed.By the protoplast fusion of S7and K15as parent strains and via orthorhombic tests‚the conditions of protoplast fu-sion were then optimized‚in which the fusion rate was up to4.603×10-5 .It is found that the fusion strain F4can clean 87% of PVA when it is cultured into activated sludge‚which is3~4times that of ordinary activated sludge.
