邻菲咯啉-铜-氨基酸三元配合物与DNA的作用

摘要: 采用荧光光谱、电子吸收光谱、粘度测定及琼脂凝胶电泳等实验方法,研究了邻菲咯啉-铜-甘氨酸、邻菲咯啉一铜一异亮氨酸和邻菲咯啉-铜-蛋氨酸三种配合物与DNA的相互作用.荧光、电子吸收及粘度实验均表明,三种配合物与DNA的作用都为部分插入;琼脂凝胶电泳显示,三种配合物对pBR322 DNA有较明显的切割作用.此外,辅助配体氨基酸空间位阻过大会阻碍配合物与DNA的插入作用,过小也不利于插入作用.

Abstract:

The interactions between DNA and three complexes,namely,[Cu(phen)(L-Gly)(H2O)]•CLO4•2.5H2O(phen=phenanthroline,L-Gly=L-Glycine),[Cu(phen)(L-Ile)(H2O)](CLO4)(L-Ile=Isoleucine)and[Cu(phen)(L-Met)(H2O)](CLO4)(L-Met=L-Methionine),were investigated by means of the nu0rescence
spectroscopy,the electronic absorption spectroscopy,the viscosity measurement and the agarose gel electrophore.sis.It is revealed by the fluorescence spectroscopy.the electronic ab sorption spectroscopy and the viscosity men.surement that the mode of the interaction between the complexes and DNA is partial intercalation.Moreover,the results of agarose gel electrophoresis indicate that all the complexes exhibit obvious pBR322 DNA cleavage activitv.Furthermore,a moderate size of the auxiliary ligand should be taken into consideration because a larger or snlaller steric hindrance of the auxiliary ligand would afect the interaction between the three designed complexes and DNA.
