IBU-PHBV纳米粒的制备、逐层自组装包覆及体外释放

Fabrication,LBL Self-Assembly and In-Vitro Release of Ibuprofen-PHBV Nanoparticles

摘要: 用溶剂蒸发法制备了布洛芬(IBU)-羟基丁酸和羟基戊酸共聚物(PHBV)纳米粒,粒径在500~800nm之间.研究了乳化剂类型及浓度、水相pH值、药/聚合物质量比、共聚物中羟基戊酸单体(HV)含量对载药纳米粒包封率和载药量的影响.最佳工艺条件为:水相pH值为4.0,药/聚合物质量比为10/50,聚合物中HV含量为6%~10%(质量分数),聚乙烯醇浓度为l%(体积分数).用逐层(LbL)自组装技术将壳聚糖和海藻酸钠包覆到载药纳米粒表面,包覆6层聚电解质后的IBU-PHBV纳米粒的突释量由包覆前的70%下降到15%,说明将溶剂蒸发法和LbL自组装技术结合可以显著抑制释放初期的药物突释效应.

Abstract:

Ibuprofen-poly(3-hydroxybutyrate-co-3-hydroxyvalerate)nanoparticles with the diameter of 500 to 800 nm were fabricated by means of the solvent evaporation method. The effects of the type and concentration of the emulsifier,the pH value of the water phase,the mass ratio of drug to polymer and the hydroxyvalerate(HV)con-tent of the polymer on the encapsulation efficiency and drug loading of the nanoparticles were also investigated. The optimized conditions were then chosen as:the pH value of water phase 4.0,the mass ratio of drug to polymer 10:50,the HV mass fraction 6% to 10%,and the polyvinyl alcohol concentration 1%(volume fraction). Moreo-ver,chitosan and sodium alginate were absorbed on the IBU-PHBV nanoparticles via the layer-by-layer(LbL)selfa-ssembly technique,resulting in a decrease in the burst amount of nanoparticles from 70% to 15% respectively be-fore and after coating polyelectrolyte in 6 layers,thus coming to the conclusion that the combination of the solvent evaporation and the LbL self-assembly is an effective way to overcome the initial burst release of drugs.
