摘要

采用稀盐酸水解玉米麸皮得到低聚糖阿魏酸酯(FOs),经D301大孔树脂的除杂和聚酰胺柱层析的分离获得FOs中的F-Ara组分,分析了其纯度和结构,并比较了该组分与阿魏酸(FA)体外清除·OH的能力.结果表明:采用D301大孔树脂和聚酰胺柱层析法从玉米麸皮酸解物中分离得到的F-Ara纯度接近HPLC色谱纯级,其体外清除·OH能力在低浓度(2~6mmol/L)时与FA相似,在高浓度(6~10mmol/L)时则明显比FA强;当浓度为10mmol/L时,F-Ara对.OH的清除率达72.9%,而FA仅为62.4%.由此可见,F-Ara是一种天然高效的自由基清除剂.

Abstract

Feruloylated oligosaccharides(FOs) were prepared by the hydrolysis of maize bran in dilute hydrochloric acid,and the component F-Ara was obtained by means of D301 macroporous resin chromatography and poly-amide resin purification from FOs.Then,the purity and structure of F-Ara were analyzed,and the capability of F-Ara for ·OH scavenging in vitro was compared with that of ferulic acid(FA).The results show that(1) the purity of F-Ara approaches HPLC-purity level after the two above-mentioned resin separation processes;(2) at low concentration(2~6mmol/L),F-Ara is similar to FA in terms of ·OH scavenging in vitro,while at high concentration(6~10mmol/L),it is more efficient than FA;and(3) at the concentration of 10 mmol/L,the percentage of scavenging ·OH by F-Ara reaches 72.9%,while that of FA is only 62.4%.Thus,it is concluded that F-Ara is a natural and efficient free radical scavenger.
