序贯试验设计改进高效硝化菌发酵培养基

Improvement of Fermentation Medium for High-Efficiency Nitrobacteria by Sequential Experimental Design

摘要: 为了开发商品化的硝化菌产品,以去除养殖水体中的亚硝酸盐,利用序贯试验设计(包括Plackett-Burman设计、部分析因设计、最速上升法和中心组合设计等)对从虾塘中筛选得到的硝化菌的发酵培养基进行优化,得到最佳培养基组成为:NaHCO31.86g/L、NaNO22.04g/L、Na2CO30.2g/L、NaCl 0.2 g/L、KH2PO40.1 g/L、MgSO4.7H2O 0.1 g/L和FeSO4.7H2O0.01g/L.由该优化培养基培养的硝化菌使亚硝酸盐的氧化速率由初始的580.7mg/(g·d)提高至859.5mg/(g·d),且其在模拟和真实养殖水体中对亚硝酸盐的降解效果良好.

Abstract:

In order to produce commercial nitrobacteria for the removal of nitrites in aquaculture water, the sequential experimental design including the Plackett-Burman design, the factorial design, the steepest ascent method and the central composite design was used to optimize the fermentation medium of nitrobacteria isolated from a shrimp pond. The optimal composition of the medium was determined as 1.86 g/L NaHCO3, 2.04 g/L NaNO2, 0. 2 g/L Na2CO3, 0. 2g/L NaCl, 0. 1 g/L KH2PO4, 0. 1 g/L MgSO4.7H2O , and 0. 01 g/L FeSO4.7H2O. The results indicate that, by using the nitrobacteria cultured with the optimal medium, the nitrite oxidation rate increases from 580. 7mg/(g · d) to 859.5 mg/( g · d), and the nitrites in both the simulated and the field aquaculture systems can effectively be oxidized.
