嗜热厌氧梭菌直接发酵造纸污泥的基本特性

Basic Characteristics of Clostridium thermocellum Cultured in Unpretreated Paper Sludge

摘要:

为有效利用造纸污泥中的纤维素和半纤维素,研究了嗜热厌氧梭菌Clostridium thermocellum 在不同底物特别是在造纸污泥中发酵的基本特性及相应的纤维素酶和木聚糖酶基本性质. 结果表明: C.thermocellum 能够直接发酵没有经过任何预处理的造纸污泥,具有作为生物联合加工( CBP) 菌株的良好潜力; 在化学制浆污泥中其乙醇产量最高达到7. 41 g /L,较商业纤维素中的乙醇产量提高了77. 90%,纤维素酶酶活和木聚糖酶酶活最高分别达到1. 92U/mL 和1. 30U/mL; 在脱墨污泥里其发酵受到明显的抑制,乙醇产量只有1. 31g /L,纤维素酶酶活和木聚糖酶酶活分别为1. 64U/mL 和0. 90U/mL; 纤维素酶最适温度为60℃,最适pH 值为5. 5; 木聚糖酶最适温度为70℃,最适pH 值为7. 0; 两种酶都表现出较强的金属离子适应性.

Abstract:

In order to effectively utilize the cellulose and hemicellulose existing in paper sludge,the basic characteristics of Clostridium thermocellum ( C. thermocellum) cultured in different substrates,especially in unpretreated paper sludge,were discussed,and the corresponding cellulase and xylanase were also investigated. The results show that C. thermocellum is a potential strain for combined bioprocessing because it can directly ferment paper sludge without any pretreatment,and that,when chemical paper sludge is used as the substrate for C. thermocellum,the yield of ethanol reaches 7. 41 g /L,which is 1. 779 times that of the ethanol obtained from using commercial cellulose as the substrate,and the cellulase and xylanase activities reach 1. 92 U/mL and 1. 30 U/mL,respectively. However,in recycled paper sludge,the ethanol yield decreases to 1. 31 g /L because the fermentation is inhibited,and the cellulase and xylanase activities are respectively 1. 64U/mL and 0. 90U/mL. Moreover,it is found that the optimal temperature and pH value for cellulase are 60℃ and 5. 5,respectively,while those for xylanase are respectively 70℃ and 7. 0,and that both cellulase and xylanase possess strong adaptability to metal ions.
